三黄片对糖尿病模型大鼠氧化应激水平的影响实验 被引量:1

摘要 目的观察三黄片对糖尿病模型大鼠氧化应激水平的影响,并探讨其作用机制。方法将50只Wistar大鼠随机分为2组,正常组12只,造模组38只。造模组大鼠采用链脲佐菌素(STZ)腹腔注射制备糖尿病模型,造模成功后随机选取24只大鼠分为糖尿病组、三黄片组,各12只。三黄片(10 g/kg)组给予三黄片研末灌胃,正常组、糖尿病组予等容积纯净水灌胃。灌胃8周后,观察3组大鼠血糖及体质量的变化,并检测大鼠血清及肾脏组织超氧化物歧化酶(SOD)及丙二醛(MDA)水平。结果灌胃8周后,3组大鼠血糖较灌胃前均无明显变化(P>0.05);灌胃前后糖尿病组及三黄片组血糖均高于正常组(P<0.05),且2组血糖组间比较差异均无统计学意义(P>0.05);灌胃8周后正常组大鼠体质量较灌胃前明显增加(P<0.05),糖尿病组及三黄片组较灌胃前均无明显变化(P>0.05),且2组体质量组间比较差异均无统计学意义(P>0.05);灌胃8周后,糖尿病组及三黄片组与正常组比较血清SOD均明显降低(P<0.05),MDA均明显升高(P<0.05),但三黄片组SOD降低幅度及MDA升高幅度均低于糖尿病组(P<0.05);灌胃8周后,糖尿病组及三黄片组与正常组比较肾脏组织SOD均明显降低(P<0.05),MDA均明显升高(P<0.05),但三黄片组SOD降低幅度及MDA升高幅度均低于糖尿病组(P<0.05)。结论三黄片对糖尿病模型大鼠具有明显抗氧化应激作用,可以升高SOD水平,降低MDA水平,预防糖尿病血管病变,预防动脉硬化,保护肾脏组织。 Objective To observe the effects of Sanhuang tablet on oxidative stress level in diabetes mellitus (DM) model rats and investigate its mechanism. Methods 50 Wistar rats were randomly divided into two groups, 12 in normal group,and 38 in model g/oup. The DM model rats were induced by streptozotocin (STZ) intraperitoneal injection. After model induced,24 rats selected from model group were randomly divided into DM group and Sanhuang tablet group, 12 rats in each group. Rats in Sanhuang tablet group were received Sanhuang powder by gavage. Rats in normal group and DM group were received the same volume purified water by garage. The changes of blood glucose and body mass in three groups were observed eight weeks after gavage administration, as well as the level of superoxide dismutase (SOD) and malondiadehyde (MDA) in rats' serum and kidney. Results Eight weeks after gavage, there was no signficant change on the level of blood glucose in three groups (P 〉 0.05 ). The level of blood glucose in DM and Sanhuang tablet groups before and after gavage was higher than that in normal group (P 〈 0.05 ), and there was no significant difference between DM and Sanhuang tablet groups on blood glucose (P 〉 0.05). Eight weeks after gavage, the body mass was obviously increased in normal group ( P 〈 0.05 ), the body mass had no changes in DM and Sanhuang tablet groups (P 〉 0.05 ), and there were no significant difference between two groups (P 〉 0. 05 ). Eight weeks after gavage, the level of serum SOD in DM and Sanhuang tablet groups was obviously decreased as compared with that in normal group ( P 〈 0.05 ), and the level of MDA was increased ( P 〈 0.05 ), the rangeability of SOD and MDA in Sanhuang tablet was lower than that in DM group (P 〈 0.05 ). Eight weeks after gavage, the SOD level of kidney in DM and Sanhuang tablet groups was obviously decreased as compared with that in normal group (P 〈0.05), and the level of MDA was increased (P 〈 0.05 ) , the rangeability of SOD and MDA in Sanhuang tablet was lower than that in DM group ( P 〈 0.05 ). Conclusion Sanhuang tablet has anti -oxidative stress effect on DM model rats, can increase the level of SOD, decreased the level of MDA, prevent diabetic angiopathies and arteriosclerosis,protect the kidney tissue.
