头痛宁对偏头痛大鼠c-fos和c-jun基因表达的影响 被引量:3

摘要 目的探讨头痛宁对偏头痛大鼠c-fos和c-jun基因表达的影响。方法 Wistar大鼠60只,分为生理盐水组、偏头痛模型组、盐酸氟桂利嗪组,头痛宁高、中、低剂量组,除生理盐水组外,其余各组注射硝酸甘油建立偏头痛模型,并用免疫组化方法观察c-fos和c-jun基因的表达。结果偏头痛模型组c-fos和c-jun阳性细胞数增多,与生理盐水组相比有统计学差异(P<0.01);头痛宁高剂量组和盐酸氟桂利嗪组c-fos和c-jun阳性细胞数减少,与偏头痛模型组比较有统计学差异(P<0.01);头痛宁中剂量组c-fos和c-jun阳性细胞数减少,与偏头痛模型组比较有统计学差异(P<0.05);头痛宁低剂量组c-fos阳性细胞数减少,但与偏头痛模型组比较无统计学差异;头痛宁低剂量组c-jun阳性细胞数减少,与偏头痛模型组有统计学差异(P<0.05)。结论头痛宁可以减弱由硝酸甘油诱导的致痛物质c-fos、c-jun的表达,从而抑制痛觉信息的传递。 Objective To explore the effect of Toutongning on the expression of c-fos and c-jun in the migrainous model.Methods 60 Wistar rats,in both genders,were randomly divided into 6 groups: the normal saline(NS)group,the model group,the Flunarizine Hydrochloride group,the high dose of Toutongning group,mid dose group,and low dose group.The nitroglycerin migrainous animal model was constructed with the nitroglycerin(NTG)subcutaneous injection.The expression of c-fos and c-jun was detected with the immunohistochemical method.Results c-fos and c-jun positive cells in the model group significantly increased compared with the NS group(P0.01).The expression of c-fos and c-jun in the high-dose Toutongning group and Flunarizine Hydrochloride group decreased compared with the model group(P0.01).The expression of c-fos and c-jun in the mid-dose Toutongning group decreased compared with the model group(P0.05).The expression of c-fos in the low-dose Toutongning group decreased,but there was no significant difference between the model group and the low-dose Toutongning group.The c-jun positive cell number in the low-dose Toutongning group decreased,compared with the model group(P0.05).Conclusion Toutongning can lower the expression of c-fos and c-jun induced by nitroglycerin,and can inhibit the transmission of nociceptive pain information.
