摘要 目的比较几种多糖的含量测定方法及其用不同标准品为对照的差异。方法对同一植物多糖,采用苯酚-硫酸法、蒽酮-硫酸法、DNS法以及葡萄糖、半乳糖、木糖三种标准品为对照进行含量测定,并用PMP衍生化后HPLC-DAD测定该多糖的单糖组成。结果苯酚-硫酸法用葡萄糖、半乳糖、木糖为对照的测定结果分别为60.60%、99.02%、55.86%;蒽酮-硫酸法的测定结果依次为59.16%、98.83%、127.69%;DNS法的测定结果依次为70.66%、69.45%、60.21%。结论多糖的含量测定应选取合适的单糖作为对照;苯酚-硫酸法以灵敏、受蛋白质干扰小而优;蒽酮-硫酸法需有合适的对照品为前提;DNS法以不同标准品为对照差异小而优。 Objective To compare different methods for the determination of polysaccharide content and methods with different standard substances as references. Methods To determine polysaccharide content in the same sample by Phenol-sulfuric acid method, Anthrone-sulfuric acid method and DNS method; meanwhile, to determine the composition of monosaccharides in the polysaccharide derivatized with PMP by HPLC-DAD. Results With glucose, galactose and xylose as references, the polysaccharide contents are 60.60%, 99.02%, 55.86% by Phenol-sulfuric acid method; 59.16%, 98.83%, 127.69% by Anthrone-sulfuric acid method; 70.66%, 69.45%, 60.21% by DNS method. Conclusion To determine the polysaccharide contents, the proper reference should be applied at first; the Phenol-sulfuric acid method is in favor of its sensitiveness and smaller interference from protein; the Anthrone-sulfuric acid method should be applied with the right reference; the DNS method is in favor of small differences with different references.
