柴芩承气汤对急性坏死性胰腺炎小鼠肺HSP70表达及肺病理改变的影响 被引量:9

Effect of Chaiqinchengqi Decoction on expression of lung heat shock protein 70 and lung pathological changes in acute necrotizing pancreatitis mice

摘要 目的探讨柴芩承气汤(Chai Qin Cheng Qi Decoction,CQCQD)对急性坏死性胰腺炎(acute necrotizing pancreatitis,ANP)小鼠肺热休克蛋白70(heat shock protein 70,HSP70)表达及肺病理评分的影响。方法将30只昆明小鼠随机分成对照组、ANP组及CQCQD组各10只。ANP模型采用8%L-精氨酸溶液腹腔注射,4.0mg·g-1·1h-1×2制作。CQCQD组予以CQCQD制剂0.1ml/10g灌胃,1次/2h,观察其第6h的肺HSP70表达及肺病理评分变化。结果 ANP组HSP70表达(0.93±0.03)与对照组(0.81±0.09)相比较,表达明显无明显改变(P>0.05),CQCQD组HSP70表达(1.42±0.21)与ANP组相比较,表达明显上调(P<0.01),与对照组相比较,表达亦明显上调(P<0.01)。ANP组肺病理评分(3.87±0.83)高于对照组(2.80±0.44,P<0.05),也高于CQCQD组(3.00±0.63 P<0.05),对照组肺病理评分与CQCQD组相比较,无统计学差异(P>0.05)。结论 CQCQD可明显上调小鼠肺HSP70表达,能明显减轻ANP小鼠肺的损伤。 Objective To investigate Chaiqinchengqi decoction(CQCQD)on expression of lung heat shock protein70 and lung pathological changes in acute necrotizing pancreatitis(ANP)mice.Methods 30 mice were randomized into control group ANP group and CQCQD group with 10 mice in each group.L-arginine was administered intraperitoneally twice at doses of 4g/kg at intervals of 1hour to induce the ANP of rats.All the rats in the CQCQD group were administrated the CQCQD at doses of 0.1ml/10 g at intervals of 2hours.The expression of lung heat shock protein 70 and lung pathological changes were observed.Results The expression of lung HSP70 in CQCQD group was 1.42±0.21(HSP70/β-actin)while that in the control group and ANP group were 0.81±0.09(HSP70/β-actin)and 0.93±0.03(HSP70/β-actin),respectively.The changes of expression of lung HSP70 in CQCQD group was more obvious than that in the ANP group and control group(both,P〈0.01).However,there was no significant difference between the ANP group and control group(P〉0.05).The pulmonary pathological score in ANP group was 3.87±0.83 while that in the control group and CQCQD group were 2.80±0.44 and 3.00±0.63,respectively.The pulmonary pathological score in ANP group was higher than that in the control group and CQCQD group(both,P〈0.05).However,there was no significant difference between the control group and CQCQD group(P〉0.05).Conclusion The findings of the study indicate that CQCQD can elevate the expression of lung heat shock protein 70 and reduce the damage of ANP mice lung.
