经肾动脉插管造影骨髓间充质干细胞移植对阿霉素肾病大鼠的影响 被引量:6

Effects of bone marrow mesenchymal stem cells transplantation with renal artery catheterization angiography via carotid artery on adriamycin nephropathy in rats

摘要 目的探讨经颈动脉途径肾动脉插管造影骨髓间充质干细胞(MSCs)移植对慢性阿霉素肾病大鼠肾脏的影响,为临床干细胞移植治疗慢性肾脏病及其给药途径选择提供参考。方法 50只雄性SD大鼠作为实验动物。36只采用经尾静脉注射阿霉素制作慢性肾病大鼠模型,造模后随机分为3组(每组12只):(1)阿霉素肾衰模型对照组(ADR组);(2)MSCs移植组(A组),取MSCs悬浮液经肾动脉插管造影输入大鼠右肾;(3)生理盐水组(B组),以A组同样方法将生理盐水输入大鼠右肾。2只用于骨髓MSCs的制备。其余12只作为空白对照组(N组)。干细胞移植后第1天和第7天分别测定各组大鼠肾功能(尿素氮、肌酐)和血红蛋白含量,第7天光镜下观察各组大鼠肾脏病理改变。结果 (1)ADR组、A组、B组与N组比较血尿素氮、肌酐明显升高(P<0.05),血红蛋白含量显著降低(P<0.05)。(2)第1天,与ADR组比较,A组、B组血尿素氮、肌酐和血红蛋白含量无统计学差异(P均>0.05);第7天时,与ADR组比较,B组血尿素氮、肌酐和血红蛋白含量无统计学差异(P均>0.05),A组血尿素氮、肌酐明显降低(P均<0.05),血红蛋白含量明显升高(P<0.05)。(3)光镜下大鼠肾脏病理改变,B组肾小球、肾小管、肾间质损伤程度与ADR组相当,A组肾间质炎症改变程度轻且范围小。结论移植MSCs有利于慢性阿霉素肾病大鼠肾脏损伤的修复,经颈动脉肾动脉插管造影是可行的途径。 Objective To investigate the impact of bone marrow mesenchymal stem cells( MSCs) transplantation with renal artery catheterization angiography via carotid artery on the kidney in rats with chronic adriamycin nephropathy in order to provide theory evidence for the clinical treatment of chronic nephropathy using stem cells transplantation and its medication approach. Method Fifty male SD rats were used as experimental animals. Thirty-six rats were used to establish chronic adriamycin nephropathy model by injecting adriamycin via caudal vein,and the modeled rats were randomly divided into three groups( n = 12 each): adriamycin-induced renal failure model control group( ADR group),MSCs transplantation group( A group) and normal saline group( B group). Two rats were used to prepare MSCs. The remained 12 rats were designed as blank control group( N group). In A and B groups,the MSCs suspension or normal saline were respectively given after renal artery catheterization angiography through carotid artery. The renal functions including serum urea nitrogen and creatinine and the hemoglobin content at 1-and 7-day after stem cells transplantation were respectively detected in all groups,and the renal pathological changes at 7-day after stem cells transplantation were observed under the light microscope in all groups. Results Compared with N group,serum urea nitrogen and creatinine significantly increase,and hemoglobin content significantly decrease in ADR,A and B groups( all P〈0. 05). Compared with ADR group,the urea nitrogen,creatinine and hemoglobin content at 1-day in A and B groups and at 7-day in B group remained all unchanged( all P〈0. 05),but the urea nitrogen,creatinine significantly decreased,and the hemoglobin content significantly increased at 7-day in A group( all P〈0. 05). The renal pathological changes under the light microscope showed that the injury degrees of glomerulus,renal tubules and renal interstitium in B group were equivalent to ADR group,but the change degrees of renal interstitial inflammation were slighter,and the inflammation range was smaller in A group compared with ADR group. Conclusion MSCs transplantation might contribute to the repairing of renal damage in rats with chronic adriamycin nephropathy,and the renal artery catheterization angiography via carotid artery is a feasible approach.
