摘要
目的研究去甲斑蝥素(NCTD)体外抑制人胰腺癌细胞株PANC-1的增殖作用。方法以不同浓度NCTD处理PANC-1细胞24 h后,采用MTT法检测其对PANC-1细胞的生长抑制作用;Hoechst 33258染色观察PANC-1细胞形态的变化;Western blot印迹法检测NF-κB p65和AKT蛋白的表达。结果NCTD可导致处理的细胞增殖速度减慢,且抑制效应随药物剂量的增加而增强,细胞出现典型凋亡形态学改变,NF-κB p65和AKT蛋白的表达均出现明显下降(P<0.01)。结论去甲斑蝥素能明显抑制胰腺癌PANC-1细胞的生长并诱导其凋亡,其机制可能是通过下调NF-κB p65和AKT的表达来实现的。
【Objective】To find out the effect of norcantharidin on the growth ability of pancreatic cancer cell line PANC-1 in vitro. 【Methods】Cell proliferation was determined by MTT assay. Apoptosis was observed by Hoechst33258 staining and expressions of NF-κB p65 protein and AKT protein were tested using Western blot.【Results】The inhibition on PANC-1 growth was increased with increasing concentration of norcantharidin. Distinct morphological changes of apoptosis were found in norcantharidin-treated cells under inverted light microscope. The expressions of NF-κB p65 protein and AKT protein were significantly reduced(P〈0.01).【Conclusions】Norcantharidin could significantly inhibit the growth of pancreatic cancer PANC-1 cells, which was associated with down-regulating the expressions of NF-κB p65 protein and AKT protein.
