TNF-α诱导人蜕膜细胞凋亡基因表达谱的分析

摘要 目的:研究TNF-α诱导人蜕膜细胞异常凋亡后基因表达谱的变化,为了解病理妊娠蜕膜细胞基因表达及药物治疗研究提供基础。方法:体外常规进行人蜕膜细胞培养,选用适宜浓度的TNF-α诱导建立人蜕膜细胞异常凋亡模型,采用人全基因组17Kc DNA基因芯片分析TNF-α作用后的基因表达谱变化情况,分析基因的变化趋势。结果:TNF-α诱导蜕膜细胞后检出基因9 320条,上调基因36条,下调基因48条。表达上调的已知基因主要为细胞周期依赖性蛋白激酶和抑制剂基因、DNA损伤及修复基因,另外还有氧化压力和炎症损伤基因等。表达下调的基因与细胞营养、细胞生长、细胞周期及信号转导相关。结论:TNF-α诱导蜕膜细胞异常凋亡。 Objective: To research the change of expression spectrum of apoptosis genes in human decidual cells induced by tumor necrosis factor-α (TNF-α), provide a basis for understanding gene expression in decidual cells of cases with pathological pregnancy and drug therapy researches. Methods : Human decidual cells were routinely cultured in vitro, suitable concentration of TNF-α was used to in- duce and establish abnormal apoptosis model of human decidual cells, human whole genome 17KcDNA gene chip was used to analyze the change of expression spectrum of apoptosis genes after TNF-α treatment, the change trends of the apoptosis genes were analyzed. Results: After inducing human decidual cells with TNF-α, 9 320 genes were detected, including 36 up-regulated genes and 48 down-regulated genes. The known up-regulated genes included cyclin-dependent kinase and inhibitor genes, DNA damage and repair genes, and oxidative stress and inflammatory lesions genes. The down-regulated genes were correlated with cell nutrition, cell growth, cell cycle, and signal transduction. Conclusion: TNF-α can induce abnormal apoptosis of human decidual cells.
