卵巢癌组织中Wnt信号通路活性检测及肿瘤恶性生物学行为的评估 被引量:8

摘要 目的:研究卵巢癌组织中Wnt信号通路活性及其与及肿瘤恶性生物学行为的关系。方法:将2012年3月~2014年5月我院收治的卵巢癌和卵巢良性病变患者纳入研究,收集卵巢癌组织和卵巢良性病变组织,分别采用荧光定量PCR和免疫印迹法检测Wnt、β-catenin、巨噬细胞移动抑制因子(MIF)、缺氧诱导因子-1(HIF-1)、血管内皮生长因子(VEGF)、β-N-乙酰葡萄糖胺基转移酶(OGT)、CD44v、整合素β1(IGTβ1)的mRNA含量和蛋白含量。结果:(1)Wnt信号通路:卵巢癌组织中Wnt、β-catenin的mRNA和蛋白含量均高于卵巢良性病变组织;(2)血管新生相关细胞因子:卵巢癌组织中MIF、HIF-1、VEGF的mRNA和蛋白含量均高于卵巢良性病变组织;(3)细胞黏附和侵袭相关分子:卵巢癌组织中OGT、CD44v、IGTβ1的mRNA和蛋白含量均高于卵巢良性病变组织;(4)MIF、HIF-1、VEGF、OGT、CD44v、IGTβ1的蛋白含量与Wnt、β-catenin呈正相关。结论:卵巢癌组织中Wnt信号通路的活性增强、血管新生相关细胞因子和细胞黏附和侵袭相关分子含量增多;Wnt信号通路可能参与肿瘤恶性生物学行为的调节。 Objective: To study the activity of Wnt signaling pathway and its correlation with tumor malignant biological behavior in ovarian cancer. Methods: Patients with ovarian cancer and ovarian benign lesions in our hospital from 2012 March to 2014 May were enrolled, ovarian cancer tissue and ovarian benign lesion tissue were collected. Then mRNA and protein con- tents of Wnt, 13-catenin, MIF, HIF-1, VEGF, OGT, CD44v, IGT-131 were detected by fluorescence quantitative PCR and im- munoblotting assay. Results= mRNA and protent contents of Wnt, 13-catenin in ovarian cancer tissue were higher than those in ovarian benign lesions tissue, mRNA and protent contents of MIF, HIF-1, VEGF in ovarian cancer tissue were higher than those in ovarian benign lesions tissue, mRNA and contents of OGT, CD44v, IGT-β1 in ovarian cancer tissue were higher than those in ovarian benign lesions tissue. MIF, HIF-1, VEGF, OGT, CD44v, IGT-β1 protein contents were positively correlated with Wnt, β-catenin contents. Conclusion: Activity of Wnt signal pathway is increased in ovarian cancer tissues, angiogenesis related cytokines and invasion and adhesion related molecule contents are upregulated. Wnt signaling pathway may be involved in the biological behavior of malignant tumor.
