摘要
目的建立HPLC法同时测定大鼠血浆中高车前苷、高车前素浓度的方法,初步探索高车前苷口服吸收规律。方法以大豆苷元为内标,采用液-液2次萃取处理大鼠血浆样品。采用AgilentCT-C18(250mm×4.6mm,5μm)和汉邦C18预柱(10mm×4.6mm,5μm);流动相:乙腈∶0.1%磷酸水溶液(25∶75);检测波长:335nm;流速:1.0mL·min^-1;柱温:30℃。以150mg·kg^-1剂量灌胃给药高车前苷,测定不同时间点大鼠体内高车前苷、高车前素的血药浓度。结果内源性杂质对样品测定无干扰。高车前苷在0.0433-2.163nmol·mL^-1,高车前素在0.333-16.65nmol·mL^-1范围内线性关系良好。血浆中高车前苷和高车前素的绝对回收率分别为82.41%-84.27%、91.00%-93.41%,方法回收率分别为97.58%-104.46%和95.41%-97.60%,日内精密度(RSD)均小于5.18%,日间精密度(RSD)均小于7.84%。样品冻融稳定性良好。灌胃给药高车前苷后,药物主要以高车前素的形式被吸收,高车前素的曲线下面积(AUC)为高车前苷的5.4倍。结论该法快速、灵敏、准确,可用于同时测定大鼠血浆中高车前苷、高车前素的含量。口服给药高车前苷,主要以高车前素的形式被吸收。
Objective To establish the HPLC method for simultaneously determining the concentrations of homoplantaginin and hispidulin in the plasma of mice and to investigate the pharmacokinetics of homoplantaginin in rats after oral administration.Methods With soybean aglycon as the internal standard,the plasma sample was treated with simple liquid-liquid extraction before HPLC.Chromatographic separation was achieved on an Agilent CT-C18 analytical column(250 mm × 4.6 mm,5 μm) coupled with a C18 guard cartridge(10 mm × 4.6 mm,5μm)at 30 ℃.The mobile phase consisted of 25 % acetonitrile and 75 % phosphoric acid in 0.1 % solution and delivered at a flow rate of 1.0 m L·min-1.The detection wavelength was set at 335 nm.Rat plasma was sampled at scheduled time points after given a single dose of 150 mg·kg-1homoplantaginin solution intragastrically,and the concentrations of homoplantaginin and hispidulin were determined by the method mentioned above.Results There were no interference peaks co-eluting with the compounds.A good linearity was obtained for homoplantaginin in the range of 0.0433 to 2.163 nmol·m L-1and for hispidulin in the range of 0.333 to 16.65 nmol·m L-1.The absolute recovery of homoplantaginin in the plasma varied from 82.41 % to 84.27 % and that of hispidulin varied from91.00% to 93.42%,and the method recoveries of the two components were from 97.58 % to 104.46 % and from95.41 % to 97.60 %,respectively.The intra-day precision was less than 5.18 % and inter-day precision was less than 7.84 %.The freezing-thaw stability of the samples was satisfactory.After oral administration,homoplantaginin was rapidly absorbed in the form of hispidulin,and the area under curve(AUC) of hispidulin was about 5.4 times than that of homoplantaginin.Conclusion The method is rapid,highly sensitive and accurate,and can be used to simultaneously determine the concentrations of homoplantaginin and hispidulin in the plasma of mice.After oral administration,homoplantaginin is mainly absorbed in the form of hispidulin.
