中药对去势小鼠生殖系统及行为学的影响 被引量:6

摘要 目的:观察中药熟地、丹参对去势小鼠子宫及行为学的影响,评价其对更年期综合征的干预作用。方法:将40只昆明种雌性小鼠随机分为正常组、模型组、熟地组、丹参组,每组10只,除正常组外,其余各组行双侧卵巢切除术模拟雌激素缺乏引起的更年期综合征。通过强迫游泳实验观察中药对去势小鼠行为学的影响,通过子宫系数、病理形态学观察及免疫组化法观察中药对去势小鼠子宫发育的影响。结果:与正常组相比,模型组子宫系数、内膜厚度及血管密度明显减少(P<0.05)。熟地组、丹参组可改善子宫系数及内膜厚度,其中熟地组内膜间质增生较明显,丹参组血管增生较明显,具有统计学意义(P<0.05)。结论:熟地、丹参具有类雌激素样作用。去势小鼠可较好的模拟更年期综合征的临床表现,熟地、丹参对其症状具有一定的缓解作用,但其机制可能不同。其中,对子宫内膜的影响熟地可促进内膜间质细胞生长,丹参可诱导血管的生成。 Objective: To observe effects of Radix Rehmanniae Preparata and Salvia on the uterus and behavior of ovariectomized rats to evaluate interventional the effects on Climacteric Syndrome. Methods: Forty Kunming female rats were randomly divided into normal group, model group, Radix Rehmanniae Preparata group and Salviae group, with 10 rats in each group. Bilateral oophorectomy was performed to simulate Climacteric Syndrome induced.Through the forced swimming test to observe the effect of castration on behaviors of mice,Pathomorphological observation and immunohistochemical method to ovserve the effect of traditional Chinese medicine on ovariectomized mice uterus. Result: Compared with the normal group, the uterus coefficient, the endometrial thickness and the endometrial vascular density decreased significantly in the model group (P〈0.05).It is indicated that the model was built successfully. The uterus coefficient and the endometrial thickness were improved in the Radix Rehmanniae Preparata group and the Salviae group. In Radix Rehmanniae Preparata group, endometrial stromal hyperplasia was obvious, while in Salviae group, endometrial Vessels hy- perplasia was obvious and statistical significance was obtained (P〈0.05). Compared with the normal group, immobility time was prolonged in the model group and was improved in the Radix Rehmanniae Preparata group and the Salviae group. However, all of them had no statistical significance. Conclusion: Radix Rehmanniae Preparata and Salvia have estrogen-like activities. The ovariectomized mice can simulate Clinical Manifestation of Climacteric Syndrome quite well. Radix Rehmanniae Preparata and Salvia can relieve clinical symptom of Climacteric Syndrome, however their mechanisms may be different. Radix Rehmanniae Preparata can promote the growth of endometrial stromal cells, while Salvia can induce the growth of endometrial Vessels.
