摘要
目的 :观察加味三物白散对人胃腺癌SGC - 790 1细胞生长的抑制作用 ,影响细胞周期及诱导人胃腺癌SGC -790 1细胞凋亡。方法 :采用MTT法 ,观察抑制率 ;应用流式细胞仪检测加味三物白散诱导人胃腺癌SGC - 790 1细胞凋亡和对细胞周期的影响。结果 :加味三物白散对人胃腺癌SGC - 790 1细胞生长有抑制作用 ,大、中、小剂量抑制率分别是 5 5 .75 %、4 2 .6 7%和 38.6 3% ,进一步研究发现中、大剂量组诱导细胞凋亡百分率分别为 9.0 6 %、96 .32 % ,与阴性对照组相比有明显差异 (P <0 .0 1) ;此外与阴性对照组相比 ,中、大剂量组使G0 -G1期细胞明显减少 ,G2 -M期明显增高 ,S期明显减少 ;小剂量组使G0 -G1期细胞明显增高 ,G2 -M期明显减少 ,S期减少。结论 :加味三物白散能抑制人胃腺癌SGC - 790 1细胞生长 ;其中、大剂量组可以明显诱导人胃腺癌SGC - 790 1细胞凋亡 ;并且加味三物白散对人胃腺癌SGC - 790
Objective:To study Modified San Wu Bai San(MSWBS)in inhibiting effect on stomach gland cancer cells SGC-7901,inducing apoptosis of stomch gland cancer cells SGC-7901 and the influcence on cell cycle.Method:MTT method was used to observe the inhibiting effect on cells.The effect of MSWBS on DNA synthesis and Cell cycle in stomach gland cancer SGC-7901 cells were investigated with the help of flow cytometry.Results:The inhibiting rates of large,medium and small concentration were 55.75%,42.67%t 38.632%,The apoptosis percents with MSWBS at moderate and large concentration were 9.06% and 96.3% respectively (P<0.01),and MSWBS had no function of inducing cancer cell differentiation.It couldn't shift the cell cluster into the G0F-G2 stage.Conclusion:MSWBS obviously inhibits the growth of stomach gland cancer SGC-7901 cells,induces SGC-7901 cells apoptosis at moderate and large concentration and influence cell cycle.
