基于不同剂量比的巴豆配伍桔梗“减毒”作用机制探讨 被引量:4

Investigation of Mechanism of Toxicity Reduction in Compatibility of Crotonis Fructus with Different Dosage Platycodon Grandiflorum

摘要 目的:探索巴豆配伍桔梗是否具有"减毒"作用及其作用机制。方法:以动物死亡状态及半数致死量(LD50)为评价指标,比较巴豆配伍不同剂量比的桔梗前后的动物中毒情况;通过测定比较巴豆配伍桔梗的不同剂量比下巴豆苷含量,从化学成分角度揭示巴豆配伍桔梗"减毒"作用机制。结果:巴豆配伍不同剂量比桔梗,其毒性反应均减小,且以1∶3剂量比值下LD50及其95%可行限抬高最多;同时配伍不同剂量比桔梗后巴豆苷含量呈现"抛物线"式变化,在1∶3剂量比时巴豆苷含量降低最多(32.57%)。结论:巴豆配伍桔梗(1∶3)剂量比值为其减毒作用最佳比值,可能与其减少了游离态巴豆苷含量有关。 Objective:To explore whether there is the effect of Platycodon grandiflorum on the reduction of the acute toxicity of Crotonis Fructus,and the mechanism of this effect.Method: The death condition and LD50 were used to be the index to evaluate the toxic conditions in the compatibility Crotonis Fructus with different dosage of P.grandiflorum.Furthermore,we revealed the mechanism of toxicity reduction via determining the content of crotonoside in the compatibility Crotonis Fructus with different dosage of P.grandiflorum.Result: The toxicity of Crotonis Fructus was reduced after the compatibility of P.grandiflorum,especially when dosage ratio between Crotonis Fructus and P.grandiflorum is 1∶3,the degree of LD50 and the 95% confidence level elevation was biggest.Besides,after the compatibility of P.grandiflorum,the content of crotonoside was changed as parabola,which elevated firstly and reduced then.The result showed the content of crotonoside reduced 32.57%,with masimum degree,when the dosage ratio is 1∶3.Conclusion: 1∶3 of the dosage ratio is the best ratio for reducing the toxicity of Crotonis Fructus in the compatibility of P.grandiflorum.And the effect may relate to the reduction of free-state crotonoside.
