淫羊藿苷对人乳腺癌MCF-7细胞增殖、凋亡作用的实验研究 被引量:10

摘要 目的:研究淫羊藿苷(icariin,ICA)对MCF-7细胞增殖/凋亡作用的影响及其作用机制。方法:采用MTT比色分析法及Annexin V/PI标记流式细胞仪检测ICA对MCF-7细胞的增殖与凋亡作用的影响,采用Western blot检测蛋白表达,研究ICA是否通过ERK/MAPK信号转导通路起作用。结果:与对照组比较,MCF-7细胞培养24 h后,ICA各组OD值均降低,差异有统计学意义(P<0.05);培养48 h后,各组差异均有统计学意义(P<0.05);培养72 h后,各组差异均有统计学意义(P<0.05)。与对照组比较,培养24 h后,10 m M ES单独作用后对MCF-7细胞无明显的增殖作用,ES加ICA组有明显的抑制作用。20 m M ES诱导结果与10 m M的结果一致。不同浓度的ICA作用48 h后,总ERK1/2和MEK蛋白表达均无明显变化,MEK磷酸化水平同样无明显变化,ERK1/2磷酸化水平增高。结论:淫羊藿苷对雌激素阳性人乳腺癌细胞有明显的抑制增殖与诱导凋亡的作用,同时可拮抗雌激素对乳腺癌细胞的增殖作用,但是其作用机制尚不清楚。 Objective: To approach the effect and mechanism of icariine(ICA) on hyperplasia and apoptosis of human breast cancer MCF-7 cell. Methods: We used colorimetric analysis and Annexin V / PI indication flow cytometry to study the influence of ICA on the hyperplasia and apoptosis of human breast cancer MCF-7 cell. And Western blot was used to detect the albumen expression. Results: When MCF-7 cell was cultured for 24 hours and compared the control group,the OD values of all the ICA groups were degraded and the difference had statistical significance(P〈0. 05). When MCF-7 cell was cultured for 48 hours,the difference of the groups had statistical significance(P〈0. 05). Compared the control group,10 m M ES had no hyperplasia effect on MCF-7 cell and ES and ICA had conspicuous depressant effect on MCF-7 cell. The derivation effect of 20 m M ES and 10 m M ES was identical. After different concentrations of ICA affecting for 48 hours,super albumen expressions of ERK1 /2 and MEK were not markedly changed and the MEK phosphorylation level was not markedly changed and the ERK1 /2 phosphorylation level was increased. Conclusion: ICA has the evident hyperplasia and apoptosis effect on MCF-7 cells. And it can inhibit the hyperplasia of ES on MCF-7 cell,but the mechanism of action is unknown.
