摘要
程序性死亡因子1(programmed cell death-1,PD-1)是属于共刺激分子28(costimulatory molecule 28,CD28)/细胞毒性T淋巴细胞相关抗原4(cytotoxic lymphocyte associated antigen,CTLA-4)家族的抑制性受体。因为PD-1的免疫抑制作用,对其机制和应用的研究一直是免疫学研究的焦点之一。如今,它对淋巴细胞的影响和作用机制已经基本清晰,而利用其研发的抗体药物也在癌症治疗中进入临床试验阶段。本文主要介绍现今已掌握的PD-1信号对淋巴细胞的作用机制,以及其在慢性病毒感染、自身免疫病和癌症治疗中的应用。
Programmed cell death (PD)-I is an inhibitory receptor that belongs to the costimulatory molecule 28(CD28)/cytotoxic lymphocyte associated antigen (CTLA-4) family. Recent evidence has highlighted the pivotal role of the PD-1 in maintaining the immunosuppressive microenvironment. Its effect on lymphocytes has been basically cleared, and antibody drugs which target this pathway also entered the stage of clinical trials in cancer treatment. In this review, we focus on the physiological role exerted by PD-1 in regulating the function of lymphocyte and its application in treating diseases such as cancer, autoimmunity disease.
