摘要
目的观测大鼠-骨骼肌损伤后不同时间点的病理学变化,为损伤时间推断的研究和应用提供依据和参考。方法 SD大鼠40只随机分为实验组(35只)和对照组(5只),实验组用500g重的打击器打击大鼠右下肢,建立大鼠骨骼肌机械性损伤动物模型,并随机均分为伤后6h、12h、1d、3d、7d、10d、14d组。应用HE染色、Mallory染色和图像分析法观测大鼠骨骼肌损伤的病理学改变。结果伤后6h,损伤区骨骼肌变性、间质大量出血和炎细胞浸润;12h^1d,骨骼肌坏死,伴大量炎细胞浸润,以PMNs和MNCs为主,数量分别达到高峰;3d起,坏死骨骼肌被逐渐清除,PMNs和MNCs数量逐渐减少,FBCs和新生骨骼肌开始出现,少量胶原纤维形成;7d,FBCs和新生骨骼肌数量继续增多,伴新生血管生成和胶原纤维沉积;10d^14d,FBCs数量和胶原纤维沉积分别达到高峰。结论大鼠骨骼肌损伤区PMNs、MNCs和FBCs数量,以及胶原纤维沉积随损伤经过时间的规律性变化,可为损伤经过时间推断研究提供参考。
Objective To observe pathological changes in different post-traumatic intervals after rats skeletal muscle injury and to provide reference for investigating skeletal muscle wound age determination in future. Methods A total of 40 Sprague-Dawley male rats were divided into control and injured groups (6h, 12h, ld, 3d, 7d, 10d and 14d after injury). We established a standardized animal model of skeletal muscle contusion in rats, which was controllable and reproducible well using a self-designed mechanical weight-drop (500g) device. A single impact was delivered to the site of the fight posterior limb. Pathological changes after rats skeletal muscle injury were investigated by hematoxylin-eosin (HE) staining, Mallory staining and image analysis. Results Degeneration, hemorrhage, and inflammatory infiltration were present in contused skeletal muscle at 6h post-injury; neerotized skeletal musele fibers were infiltrated by a large number of inflammatory eells, especially PMNs and MNCs. The number of PMNs and MNCs infiltrated into injured sites reached respectively a peak at 12h ld post-injury; from 3d post-injury onward, neerotized skeletal muscle fibers were phagoeytosed gradually. And the number of PMNs and MNCs was decreased gradually. FBCs, collagen fibers and regenerating myofibers were present in injured sites; the number of FBCs and regenerating myofibers augmented remarkably at 7d post-injury. Simultaneously, angiogenesis and collagen fibers were also observed; at 10 - 14d post-injury, the number of FBCs and collagen fibers deposition reached respectively a peak. Conclusion The number of PMNs, MNCs, FBCs, and the percentage of collagen fibers deposition in skeletal muscle injured zones was respectively time-dependent changes, which may provide reference for wound age determination in forensic medicine.
