摘要
目的探讨结肠癌组织中IGF-1及其亚型和Ki-67在结肠癌(CRC)组织中表达的意义及相关性。方法以确诊CRC的80例患者为研究对象,术中收集肿瘤及非肿瘤组织制备标本并分别归入观察组与对照组。行免疫组化法评估IGF-1及其亚型蛋白和Ki-67抗原的表达。统计分析两者表达与临床资料间的关系。结果 1观察组IGF-1、IGF-1A、IGF-1B蛋白及Ki-67抗原表达的标志指数(LI)显著高于对照组(P<0.05),但IGF-1C蛋白表达LI无显著差异;IFG-1与IGF-1A表达呈正相关、与Ki-67呈负相关,IGF-1B及IGF-1C与Ki-67呈正相关,上述相关性有统计学意义(P<0.05),未见其他显著相关性。2IGF-1及其亚型的表达与患者年龄呈正相关;IGF-1C及Ki-67的表达与肿瘤分化程度呈负相关;IGF其亚型和Ki-67的表达与Dukes分期呈正相关;Ki-67表达与淋巴结转移呈正相关。上述相关性均有统计学意义(P<0.05),未见其他显著相关性。结论 IGF-1亚型与Ki-67的表达存在一定相关性,而Ki-67表达上调与结肠癌的发展有关,因此调控IGF-1基因表达可成为结肠癌治疗的新靶点。
Objective To discuss the expression meaning and correlation of IGF-1 subtypes and Ki-67 in colorectal cancer (CRC) tissues. Methods Eighty cases of patients with a definite diagnosis of CRC were selected as the research objects, tumor and non tumor tissue specimens of intraoperative collection were prepared and were divided into obser- vation group and control group. The expression of the subtypes of IGF-1 protein and Ki-67 antigen were assessed by immunohistochemistry method. The relationship between expression and clinical data were statistically analyzed. Results ①IGF-1, IGF-1A, IGF-1B protein and Ki-67 antigen expression index (LI) of observation group were significantly higher than those of control group (P〈0.05),but there was no significant difference in IGF-1C protein expression of LI; IFG-1 and IGF-1A expression had positive Correlation, negative correlation with Ki-67, IGF-1B & IGF-1C and Ki-67 had positive correlation, the correlations above were statistically significant (P〈0.05), there were no other significant correlations. ②The expression of IGF-1 and its subtypes had positive correlation with patient ages; IGF-1C and Ki-67 expression had negative correlation with tumor differentiation degree; Expression of IGF subtypes, Ki-67 and Dukes stages were positively correlated; The expression of Ki-67 was positively correlated with lymph node metastasis. The correlations above were statistically significant (P〈0.05), there were no other significant correlations. Conclusion There are correlations between the IGF-1 subtypes and the expression of Ki-67, The Ki-67 expression up is related to the development of colorectal cancer, therefore regulating and controling IGF-1 gene expression may become a new target treatment for colorectal cancer.
