Wnt/β-catenin通路相关蛋白在复发胶质瘤中的表达及其临床意义 被引量:6

Expression and Clinical Significance of the Members of Wnt/β-catenin Signaling Pathway in Recurrent Gliomas

摘要 目的探讨低级别和高级别复发脑胶质瘤中Wnt/β-catenin通路中相关蛋白β连接素(β-catenin)、磷酸化糖原合成激酶3β(p-GSK-3β)、结肠腺瘤性息肉病蛋白(APC)和轴抑制蛋白(Axin)的表达及其临床意义。方法采用免疫组化方法检测10例正常脑组织和30例各级别复发胶质瘤标本中β-catenin、p-GSK-3β、APC及Axin蛋白的表达。结果β-catenin蛋白在正常脑组织中不表达,在复发胶质瘤组织的阳性表达率为100%(P<0.01);p-GSK-3β蛋白、APC蛋白、Axin蛋白在正常脑组织中表达率(分别为90%、100%、100%)高于在复发胶质瘤组织的阳性表达率(分别为26.7%、13.3%、10%)(P<0.01);β-catenin在复发低级别胶质瘤和高级别胶质瘤中的阳性表达率均为100%;p-GSK-3β、APC、Axin蛋白在复发低级别胶质瘤阳性表达率(分别为35.7%、28.6%、21.4%)高于在复发高级别胶质瘤中的阳性表达率(分别为18.8%、0%、0%)(P<0.05)。结论β-catenin、GSK-3β、APC、Axin蛋白异常表达与复发脑胶质瘤发生密切相关,且磷酸化GSK-3β、APC及Axin蛋白低表达或不表达与胶质瘤恶性程度有关。 Objective To investigate the clinical significance and expression of the members of the Wnt/β - eatenin signaling pathway including β- catenin, p - GSK - 3 β ( glycogen synthase kinase - 3 β ), APC ( adeno- matous polyposis coli protein )and Axin (axin inhibition protein )in various fade of recurrent glioma. Methods Immunohistochemistry was used to determine the expression of β - catenin, GSK - 3β, APC and Axin protein in 30 samples of recurrent gliomas and 10 samples of normal brain tissues respectively. Results There was no expression of β - catenin protein in normal brain tissue. The expression rate of β - catenin protein in low - grade recurrent gliomas and high - grade recurrent gliomas was 100% (P 〈0.01 ). The expression of p -GSK -3β ,APC ,Axin protein in normal brain tissue were 90% , 100% and 100% while they were 26.7% ,13.3%, 10% in low- grade gliomas and high- grade recurrent gliomas (P 〈 0.01 ). The ex- pression of β - catenin protein in low - grade gliomas and high - grade recurrent gliomas all was 100%. The expression of p -GSK -3β in low -grade recurrent gliomas and high -grade recurrent gliomas were 35.7% , 18.8% (P 〈 0.05 ). The expression of APC, Axin protein in low - grade recurrent gliomas were 28.6% , 21.4% while they all were 0% in high -grade recurrent gliomas( P 〈 0.05 ). Conclusion The abnormalities of expression β -catenin,p- GSK- 3β ,APC and Axin are closely related to the tumorgenesis. Meanwile,the low - expression or none - expression of p - GSK - 3 β, APC and Axin APC be relevant to the grade malignancy of recurrent gliomas.
