摘要
目的探讨丹参酮ⅡA提取工艺及其诱导SGC7901胃癌细胞凋亡机制。方法正交实验方法优化丹参酮ⅡA提取工艺,用MTT法及电镜观测其对SGC7901胃癌细胞凋亡的影响。结果丹参酮ⅡA对SGC7901胃癌细胞增殖有明显的抑制作用,并呈现浓度和时间的依赖性。8.0μg/mL丹参酮ⅡA作用72 h后,电镜下出现凋亡小体。流式细胞仪检测丹参酮ⅡA 2.0、4.0和8.0μg/mL作用72 h后细胞生长抑制率为(25.70±1.86)%、(47.80±2.75)%、(58.40±3.25)%。结论丹参酮ⅡA最佳提取工艺为:加10倍量的80%乙醇,提取1次,提取时间为45分钟;丹参酮ⅡA能诱导SGC7901胃癌细胞凋亡。
[Objecttive] To study the optimum extraction process for Tanshinone Ⅱ A and its mechanism of inducing apoptosis of gastric cancer cell line SGC7901. [Methods] Optimize the extraction process for Tanshinone Ⅱ A by orthogonal experimental and using MTT method and electron microscope to observe its' effect of apoptosis of human gastric carcinoma cell SGC7901. [ Results ] Tanshinone Ⅱ A has significant inhibition effect on cell proliferation of gastric cancer cell SGC7901 with concentration and time dependence. 72 h after 8 μg/mL Tanshinone Ⅱ A impact on cancer cells, apoptotic bodies were observed under electronic microscope. Cell growth inhibition rate was (25.7±1.86)%, (47.8±2.75)%, (58.4±3.25)% detected by flow cytometry method after 72 h of Tanshinone ⅡA in dosage of 2, 4 and 8μ g/mL, respectively. [ Conclusions ] The optimum extraction technology of Tanshinone Ⅱ A:extract once after adding 10 times amount of 80% ethanol in 45 minutes. Tanshinone A can induce apoptosis of gastile cancer cell line SGC7901.
