中波紫外线对HaCaT细胞核因子-κB和p53表达的影响 被引量:1

摘要 目的研究中波紫外线(UVB)对HaCaT细胞核因子-κB(NF-κB)和p53表达的影响。方法以60mJ/cm2的UVB照射HaCaT细胞后8h、16h和24h,采用real time PCR和western blot等技术检测NF-κB和p53的表达情况。结果 1与未处理组相比,UVB照射HaCaT细胞8h、16h和24h后,NF-κB mRNA水平升高程度分别为27%、32%和42%,p53 mRNA水平升高程度分别为13%、20%和28%;2未处理组HaCaT细胞NF-κB和p53蛋白的表达水平分别为0.3989±0.04802和0.3018±0.03605,UVB照射8h、16h和24h后,二者的表达均随时间延长而升高,NF-κB为0.4283±0.0195、0.5976±0.0401和0.8255±0.0214,p53为0.3925±0.0244、0.4595±0.0362和0.5811±0.0482,差异有统计学意义(P<0.05)。结论 UVB可致HaCat细胞中NF-κB和p53表达水平升高。 Objective To investigate the effects of uhraviolet-B on the expression of nuclear factor-κB (NF-κB) and p53 in HaCaT ceils in vitro. Methods The expressions of NF-κB and p53 in the HaCaT cells treated with UVB of 60mJ/cm^2 were assessed by real time PCR and western blot at settled duration of 8h, 16h and 24h. Results Contrasted with untreated HaCat ceils, NF-κB mRNA in HaCaT cells treated with UVB increased by 27%, 32% and 42% at 8h, 16h and 24h individually, while p53 increased by 13%, 20% and 28%. NF-κB and p53 protein could be detected in untreated HaCaT cells. The expression of NF-κB and p53 time-dependently increased after the HaCaT ceils be treated with UVB (P〈0.05).Conclusion UVB could stimulate NF-κB activity and increase the expression of p53 in HaCaT cells.
