紫色悬钩子水提物体内外抑制α-葡萄糖苷酶的活性研究 被引量:4

Study on the water extracts of Rubus irritans against α-glucosidase enzyme activity in vivo and in vitro

摘要 目的探讨紫色悬钩子(RSI)水提物在体内、体外对α-葡萄糖苷酶(AG)的抑制活性。方法利用酶-抑制剂体外筛选模型,阿卡波糖为对照品,对不同浓度的RSI进行体外酶抑制活性研究。体内进行糖耐量试验,正常小鼠分别ig RSI 40、10 g·kg^-1,阿卡波糖0.375 g·kg^-1,生理盐水0.3 mL,连续5 d,分别负荷葡萄糖2.0 g·kg^-1、淀粉2.0 g·kg^-1,在15、30、60、120 min时测定小鼠的血糖值。结果 RSI 0.0195~2.50 mg·mL^-1时,体外酶抑制率为5.20%~88.81%;阿卡波糖1.5625~25.00 mg·mL^-1时,抑制率为9.28%~64.87%;淀粉糖耐量的提高作用比葡萄糖糖耐量的明显(P〈0.01),各给药组小鼠对淀粉和葡萄糖糖耐量均有不同程度改善。结论 RSI中含有α-葡萄糖苷酶抑制剂的活性成分,体外抑制AG活性的作用强于阿卡波糖,体内可提高正常小鼠的淀粉耐糖量。 OBJECTIVE To study the internal and external inhibition of RSI against α - glucosidase (AG) enzyme activity in vivo and in vitro. METHODS The enzyme - inhibitor activity was studied by using the external screening model of the enzyme - inhibitor, Acarbose as contrast drug and the different density of Rubus irritans (RSI). The internal sugar tolevence test:40,10 g· kg^-1 of RSI, 0. 375 g· kg^- 1 of Acarbose, and 0.3 mL of NaC1 were respectively used to lavage the normal mice ig in successive 5 days, then each group of these lavaged mice was respectively loaded with 2.0 g·kg ^-1 of glucose ,and 2.0 g·kg ^-1 of starch,their blood sugar values were measured within 15, 30, 60,120 min. RESULTS In the enzyme inhibitor model, at the density between 0. 0195 - 2.50 mg· mL^-1, RSI's inhibition rate was 5.20% -88.81%, at the density between 1. 562 -25.00 mg· mL^-1, Arcabose' s inhibition rate was 9.28% -64. 87%. In the sugar tolerance test, the sugar tolerance of starch was more than that of glucose(P 〈 0.01 ), and each dosage group had improved their starch tolerance and glucose tolerance differently. CONCLUSION RSI contains the active compound activity of AGI, whose effect of external inhibition against AG is stronger than Acarbose in vitro. RSI can increase the sugar tolerance of the normal mice in vivo.
