脾气虚证代谢综合征大鼠血液代谢组学研究 被引量:10

摘要 目的:通过对脾气虚证代谢综合征大鼠模型血浆标本进行代谢组学检测,寻找特征性代谢产物,诠释代谢物与生理病理变化的相对关系,从而揭示脾虚证代谢综合征中医证本质。方法:健康雄性SD大鼠按随机数字表法分成空白对照组和模型组,劳倦加饮食不节法复制脾气虚证代谢综合征模型。运用超高压液相色谱-四极杆-时间飞行串联质谱仪技术检测2组血浆样本中的小分子代谢产物,对所得到的代谢指纹图谱进行主成分分析和偏最小二乘法-判别分析,找出具有差异的特征代谢物。结果:找到的潜在生物标志物主要有N-乙酰基-D-葡萄糖胺、溶血磷脂酰胆碱、鞘磷脂、5-甲基胞嘧啶、促黑素抑制素、三羟基异黄酮、前列腺素A2、同型半胱氨酸、丝氨酸等。结论:脾气虚证代谢综合征的发生与糖类、脂类、蛋白质代谢紊乱有关,这些体内的小分子化合物可能是代谢综合征中医证候的物质基础。 Objective: In this study, by copying the spleen deficiency syndrome metabolic syndrome rat model, we researched model plasma specimens by metabolomies and looked for spleen deficiency syndrome metabolic syndrome lesions characteristic metabolites, further revealed the essence of spleen deficiency syndrome metabolic syndrome. Methods : Spleen deficiency syndrome and metabolic syndrome rats were divided randomly into two groups : blank group ( n = 6) and model group (n = 6). We used UPLC/Q-TOF-MS technology and detected small molecule metabolites in plasma. We identified the differences in charaeteristics of metabolites and found some biomarkers. Result: We found a number of potential biomarkers related to the metabolic syndrome rat model of : N-acetyl-D-glucosamine, lysophosphatidylcholine, sphingomyelin, 5- methylcytosine, Melanostatin, Genistein, Prostaglandin A2, homocysteine, L-Homoserine. Conclusion: The collection of these substances together constitute the Spleen Deficiency Syndromemetabolic syndrome rat model of metabolic group characteristics and are the results of the carbohydrates, lipids, proteins and other metabolic disorders.
