摘要
目的探讨聚乳酸-聚三亚甲基碳酸酯/GDN导管对大鼠脊髓损伤区GAP-43蛋白表达的影响。方法通过Western Blot法从分子生物水平对GAP-43蛋白的表达进行分析,从基因水平上来检测植入PLA-PTMC/GDNF复合导管与其它治疗组GAP-43基因表达上的差异。结果术后第2周时,B组、C组和D组GAP-43蛋白表达比正常组增高;B组、C组和D组各组间GAP-43蛋白的表达水平无显著性差异。术后第4周时,D组的GAP-43蛋白表达与2周时明显增高;术后第6周时,各组GAP-43蛋白表达仍增加,但增幅不同,C组和D组与正常对照组、A组、B组相比明显增高,D组与C组相比无显著性差异;8周时,各组GA-P43蛋白的表达与6周相比均有所均下降,D组与正常对照组及A组、B组和C组相比较仍增高。结论 PLA-PTMC/GDNF复合导管较单一应用PLA-PTMC或GDNF更有利于轴突再生进而促进脊髓损伤的功能恢复。
Objective To explore the influence of PLA-Poly-PTMC with GDNF on the protein expression of GAP-43 in spinal cord injury of rats.Methods To explore the protein expression of GAP-43 at molecular biological level with Western blot to make certain the difference between PLA-PTMC/GDNF and others.Results B,C,D groups showed high protein expression at 2weeks,and there is no significant difference among these groups.D group had significant higher expression at 4weeks that that of two weeks.C,D groups showed high protein expression at 6weeks than other groups and all groups showed higher expression.At 8weeks,all groups showed lower expression of GAP-43,but D group showed more expression than others.Conclusion Conclusion The functional Recovery in the composite catheter group(Group D)function is best,the repair of the recovery of Spinal Cord Injury is superior to a single application of PLA-PTMC catheter group or a single application of GDNF group.
