摘要
为解决细叶石仙桃人工栽培种苗短缺问题,以细叶石仙桃蒴果为材料,应用植物组织培养技术,筛选不同阶段所需的最适培养基。结果表明其种子可在MS+20g·L-1蔗糖+8g·L-1琼脂培养基上萌发,发芽率可达70%以上;MS+50g·L-1土豆+20g·L-1蔗糖+8g·L-1琼脂培养基有利于原球茎分化小苗,低浓度6-BA和NAA组合适合原球茎增殖与芽的分化;在1/2MS+0.5mg·L-1 NAA+20g·L-1蔗糖+8g·L-1琼脂培养基上生根率达100%,通过以上系列培养基的培养可获得完整植株。
To alleviate a seed shortage on Pholidota cantonensis Rolfe ,artificial culturing with medium optimization was investigated by using the plant tissue culture technology .The results showed that MS+20 g · L^-1 sucrose+8 g · L^-1 agar was an appropriate medium that allowed a germination rate above 70% .MS+50 g · L^-1 potato+20 g · L ^-1 sucrose+8 g · L^ -1 agar was desirable for the plantlet′s protocorm differentiation .Low concentrations of 6-BA and NAA in medium was better for bud protocorm proliferation and differentiation .The medium formulated with 1/2 MS+0.5 mg · L^-1 NAA+20 g · L^-1 sucrose+ 8 g · L ^-1 agar was optimal for rooting ,as a rate of 100%was achieved .By using the combination of the above-mentioned media at respective stages ,the plantlets of P . cantonensis Rolfe could be rapidly obtained from seeds .
