摘要
初步探讨霉酚酸酯(MMF)治疗GVHD的分子机制。采用MMF每日1~2g,并应用RT-PCR扩增3例GVHD患者MMF使用前后外周血单个核细胞的TCR Vβ24个亚家族的CDR3,了解患者TCR VβT细胞的分布情况。结果表明,患者在GVHD发生前,有TCR Vβ3及其他亚家族基因表达,MMF治疗后,GVHD得以缓解,Vβ3不表达;当患者GVHD复发时,Vβ3基因又表达,治疗后不表达。MMF作用的分子机制可能与抑制TCR Vβ亚家族基因表达有关,TCR Vβ3可能是MMF作用的靶基因。
To approach to the molecular mechanism of treatment with Mycophenolate mofetil ( MMF ) on GVHD patients, the distribution of TCR Vβ T repertoire was determined with amplification of the CDR3 of TCR Vβ 24 subfamily genes by using RT-PCR in 3 cases with GVHD following allogeneic hematopoietic stem cell transplantation. It was found that they were expressions of TCR Vβ3 gene and other subfamily genes before GVHD, but they were not detected after MMF treatment with remission of GVHD, while during relapse of the disease, expressions of Vβ3 gene reappeared, but disap-peared after treatment. The molecular mechanism for the treatment with MMF on GVHD might be correlated with the in-hibition of TCR Vβ subfamily gene expressions and the TCR Vβ3 gene might be the target gene for MMF.
