2/100Hz电针抑制吗啡戒断大鼠中枢神经元型一氧化氮合酶的表达 被引量:2

Effect of 2/100 Hz electroacupuncture on neuronal nitric oxide synthase expression in the central nervous system in morphine withdrawal rats

摘要 目的 :观察 2 / 10 0Hz电针 (electroacupuncture,EA)对吗啡戒断大鼠中枢相关区域神经元型一氧化氮合酶(neuronalnitricoxidesynthase,nNOS)表达的影响。 方法 :给大鼠腹腔注射 (i.p .)递增量吗啡 10d ,造成大鼠对吗啡的依赖。在最后一次注射吗啡后 12和 2 4h分别给予 2 / 10 0HzEA 30min。电针结束后 30min ,取大鼠脊髓和脑 ,采用免疫组织化学方法和计算机图象分析技术 ,对脊髓腰膨大 (L4 L5)、蓝斑 (locuscoeruleus ,LC)、中脑导水管周围灰质 (periaqueductalgray ,PAG)nNOS的原位表达进行观察及半定量分析。 结果 :吗啡戒断大鼠脊髓L4 L5第X板层、LC及PAG腹外侧区的nNOS阳性神经元数量显著增多 ,L4 L5第I IV板层、LC及PAG的nNOS的免疫染色光密度 (D)明显增强 ;给予 2 / 10 0HzEA后 ,吗啡戒断大鼠上述部位的nNOS阳性神经元的数量明显减少 ,nNOS的D值显著降低。结论 :2 / 10 0HzEA能抑制吗啡戒断大鼠中枢相关区域nNOS的表达 。 Objective: To investigate the effect of 2/100 Hz electroacupuncture (EA) on the expression of the central neuronal nitric oxide synthase (nNOS) in morphine withdrawal rats. Methods: Rats were made dependent on morphine by repeated morphine injections (5-160 mg·kg -1, i.p., twice a day) for 10 days. They were then given 2/100 Hz EA for 30 min 12 and 24 hours after the last injection of morphine. Immunohistochemistry was used to detect the protein expression of nNOS in lumber spinal cord (L 4-L 5), locus coeruleus (LC) and periaqueductal gray (PAG) in morphine withdrawal rats. Results: A significant increase in the number of nNOS positive neurons and the optical density of nNOS immunostaining was observed in the L 4-L 5, LC and PAG 25 h after the last morphine injection in morphine withdrawal rats as compared with the control rats. This increase was almost totally reversed by 2/100 Hz EA stimulation. Conclusion: The increased expression of nNOS in the central nervous system of morphine withdrawal rats could be almost completely reversed by EA, which may account, at least partly, for the suppression of morphine withdrawal syndrome by the electroacupuncture stimulation.
