摘要 目的 :探讨间期核荧光原位杂交检测肺癌和癌旁支气管上皮细胞中 3p、9p丢失以及将其应用肺癌早期诊断的可行性。方法 :选取DNA特异的 3p、9p探针 ,以荧光原位杂交方法对 2 1例短期培养的肺癌和癌旁支气管上皮细胞中 3p、9p丢失进行分析。结果 :3p、9p丢失在癌旁支气管上皮细胞中为14 2 9% (3/ 2 1)、2 8 5 7% (6 / 2 1) ;在肺癌细胞中为 6 6 6 7% (14 / 2 1)、80 95 % (17/ 2 1)。 3p、9p均丢失者在肺癌细胞中占 5 7 14 % (12 / 2 1) ,在癌旁支气管上皮细胞中为 0。结论 :①间期核荧光原位杂交可以检测肺癌及癌旁上皮细胞中 3p、9p的丢失 ;② 3p、9p的丢失在肺癌细胞中是频发事件 ,明显高于癌旁支气管上皮细胞 ;③检测 3p。 Objective The aim of this study was to determine the deletion of 3p and 9p in carcioma and adjacent bronchial epithelial cells of NSCLC,and whether this techonique could be used for early diagonsis of lung cancer .Methods We analyzed the deletion of 3p and 9p in short term cultured lung cancer and adjacent bronchial epithelial cells of 21 cases by FISH with DNA specific probes.Results The deletion rates of 3p and 9p were 14 29%(3/21),28 57%(6/21) in adjacent bronchial epithelial cells and 66 57%(14/21),80 95%(17/21) in lung cancer cells;57 14%(12/21) cases had both 3p and 9p deletion in lung cancer cells,while no one in adjacent bronchial epithelial cells.Conclusions ①Interphase FISH could be used to detect the deletion of 3p and 9p in either lung cancer cells or bronchial epithelial cells.②The losses of 3p and 9p were frequent events in lung cancer cells,and it is more often than in adjacent bronchial epithelial cells.③The examination of deletion of 3p and 9p may provide new approaches to early diagnosis of lung cancer.
