摘要
目的 建立人羊膜 (HAM)负载培养骨髓间充质干细胞 (MSCs)生长的组织工程方法 ,观察MSCs生长增殖的形态特点。方法 将贵州小香猪的MSCs原代培养 ,传代扩增后 ,以不同密度多次接种于HAM基质面 ,逐日于倒置显微镜下观察MSCs的生长增殖变化情况 ,并于培养 8、1 8d后进行光镜、电镜观察。结果 接种后 30min内明显见到MSCs在HAM上贴附生长 ,MSCs能以合适的接种密度在羊膜上良好生长。结论 HAM对MSCs有明显的促增殖作用 ,MSCs可以HAM为载体在体外进行培养 。
Objective To observe the morphological characteristics of human amniotic membrane (HAM) loading mesenchymal stem cells (MSCs) derived from porcine bone marrow. Methods MSCs of Guizhou minipigs were primarily cultured and subsequently expanded and bred on the stroma surface of HAM with different densities for many times. The growth and proliferation of MSCs were observed under the invert microscope every day and under light microscope and electron microscope at d 8 and d 18 after culture. Results MSCs adhered to HAM about 30 min after ihoculation. MSCs grew fine on HAM when they were bred with an appropriate culture density. Conclusion HAM promotes the proliferation of MSCs significantly. MSCs may be cultured on HAM in vitro. HAM is a good carrier of MSCs.
