流式细胞仪检测细胞因子的方法学研究及临床应用

摘要 目的 寻求一种新的检测细胞因子的方法 ,探讨小儿T淋巴细胞性急性白血病 (T -ALL)细胞内的干扰因子IL -10和IL -12的表达及其临床意义。方法 应用流式细胞仪 (FCM)测定七例T -ALL患儿白血病细胞和正常T细胞中的IL -10和IL -12表达。结果 IL -2在T -ALL患儿非白血病T细胞中部分有表达 ,而正常对照T细胞和T -ALL患儿白血病细胞均无表达。IL -10在T -ALL患儿正常T细胞、白血病细胞和正常对照T细胞中均未检测到表达。结论 应用FCM对白血病患儿细胞内细胞因子的检测可为临床诊治提供客观的实验数据。急性淋巴细胞性白血病 (ALL)患儿体内可能存在TH1/TH2样细胞因子产生的失平衡状态 ,这可能是ALL发病中的重要机制之一。 Objective To seek a new method to detecting cytokines;To study expression and clinical significance of IL-10 and IL-12 in children with T-ALL. Methods Measurement of IL-10 and IL-12 in leudemic cells and normal T cells by FCM. Results IL-12 has positive expression in non-leukemic T cell of children with T-ALL, but no expression in leukemic blast T cell in children with T-ALL or in T cell of healthy children. There are on IL-10 positive expression in non-leukemic T cells and leukemic cells of children with T-ALL or in T cells of healthy children. Conclusion It can provid objective experiment data for clinical diagnosis and treatment through detecting intracelluar cytokines; The maladjusted state of TH1/TH2 cytokines may exist in ALL children. This may be one of the important mechanism in the occurence of ALL.
