摘要
目的 分离和鉴定日本血吸虫 (Schistosomajaponisum ,Sj)新基因。 方法 应用表达序列标签 (Expressedse quencetags ,ESTs)法随机筛选SjcDNA文库 ,通过PCR直接序列测定技术和同源性比较对阳性插入片段进行鉴定。采用亚克隆技术和生物信息学分析对日本血吸虫RNA聚合酶Ⅱ转录延长因子SⅢ -p15亚单位进行结构和功能的鉴定。结果 我们从SjcDNA文库中随机筛选到一个cDNA克隆 ,它含有一个 371个碱基组成的阅读框 ,编码 118个氨基酸 ,与哺乳动物等的RNA聚合酶Ⅱ转录因子SⅢ - p15亚单位具有较高的同源性 ,且具有该因子所特有的氨基酸序列 ,表明我们已经获得了日本血吸虫RNA聚合酶Ⅱ转录因子SⅢ - p15亚单位。 结论 获得了日本血吸虫RNA聚合酶Ⅱ转录因子SⅢ - p15亚单位的全长cDNA序列 。
Aim To isolate and identify novel genes of Schistosoma japonicum(Sj) from cDNA library Methods cDNA clones were randomly screened from Sj cDNA library by EST stratagem The inserts of positive cDNA clones were identified by PCR direced sequencing and homologous searching of ESTs,the cDNA sequences encoding Sj small subunit of RNA polymerase Ⅱ transcription factor SⅢ was analyzed by sub cloning and bioinformatics Results a cDNA clong was isolated from Sj cDNA library,which sequence included a ORF of 371 bp,and translated into 118 amino acid residues The deduced protein was similar to the corresponding sequence of other biologies Conclusion the full length cDNA sequences encoding Sj small subunit of RNA polymerase Ⅱ transcription facter SⅢ was obtained,which gave the basis for further functional study of this gene
