Cyclin E和p27^(kip1)在膀胱移行细胞癌中表达及与Ki-67表达的关系

摘要 目的 探讨细胞周期调控基因蛋白cyclinE和p2 7kip1在膀胱移行细胞癌 (BTCC)中的表达及与Ki -6 7表达的关系。方法 采用免疫组化SP法 ,观察 6 5例BTCC中三种指标的表达情况 ,并用 12例正常黏膜作对比。结果 cyclinE在BTCC中阳性表达率随着病理分级 (P <0 0 5 )、临床分期和Ki- 6 7指数 (P <0 0 1)的升高而增高 ,有淋巴结转移组非常显著高于无淋巴结转移组 (P <0 0 1) ;而p2 7kip1在BTCC中的阳性表达率随着临床分期、病理分级和Ki- 6 7指数的增高而降低 (P <0 0 5 ) ,有淋巴结转移组显著低于无淋巴结转移组 (P <0 0 5 )。结论 cyclinE对BTCC的分化、增殖、浸润和转移可能起促进作用 ,而p2 7kip1则对BTCC的分化、增殖、浸润和转移可能起抑制作用。两者在细胞周期进展中的共同表达失调 ,可能是导致BTCC发生发展和向恶性表型转化的机制之一。联合检测cyclinE、p2 7Kip1和Ki- 6 7的表达对BTCC的生物学行为评价和判断预后具有重要意义。 Objective To investigate the expression of cell cycle regulating protein cyclin E and p27 kip1 in bladder transitional cell carcinoma (BTCC) and to evaluate its relation with the expression of Ki-67.Methods With immunohistochemistry assay , the expressions of cyclin E , p27 kip1 and Ki-67 were examined in 65 cases of BTCC and 12 cases of normal bladder tissue as controls.Results There was a positive correlation between cyclin E expression and the pathological grade(P<0 05) , clinical stage , lymphoid node metastasis of BTCC and Ki-67 labeling index (P<0 01), and there was a negative correlation between p27 kip1 expression and the four indexes(P<0 05) . Conclusions Cyclin E may play a promoting role in the differentiation , proliferation, infiltration and metastasis of BTCC ;As contrast to cyclinE, p27 kip1 may play an inhibitive role in it . The disregulation of coexpression of the both cell cycle regulators may be one of the mechanisms of BTCC development and transformation to malignant phenotype. Examinations of cyclin E , p27 kip1 and K-67 expression in BTCC may have an important significance to judge its prognosis and biological behavior .
