摘要 目的通过检测XIAP在骨肉瘤组织中的表达,探讨其对骨肉瘤发生、发展的可能作用及临床意义。方法应用免疫组化SP法检测31例骨肉瘤化疗前穿刺标本、10例正常骨组织和10例骨软骨瘤手术切除标本XIAP的表达,并分析XIAP表达与各临床病理参数的关系。结果XIAP在正常骨组织、骨软骨瘤及骨肉瘤组织中均有表达。正常骨组织和骨软骨瘤组织都呈低表达或不表达,而骨肉瘤组织普遍高表达,较前二者均有显著性差异(P=0.000)。31例骨肉瘤组织中,XIAP表达强度与性别、年龄、肿瘤部位、肿瘤大小、肿瘤术前化疗坏死率等无显著相关(P>0.05)。结论XIAP在骨肉瘤的发生、发展中起重要作用,XIAP表达可作为骨肉瘤生物学行为评价的独立危险因素。 Objective To investigate the correlation of the expression of XIAP, with the pathogenesis and development of osteosarcoma, and its clinical significance. Methods The expression of XIAP was detected by SP immunohistochemistry in 31 biopsy specimens obtained before chemotherapy of osteosarcoma, as well as 10 specimens of normal bone tissue, and 10 speicimens of benign bone tumor (osteochondroma) from surgical resection. Then we analysed the relationship between XIAP expression and the clinical and pathological features of osteosarcoma. Results Low or scarce XIAP expression was found in normal bone tissues and osteochondroma, whereas high expression found in osteosarcoma. The difference was significant (P=0.000). In the 31 specimens of osteosarcoma, XIAP expression was not related to the patients'gender, age, tumor location, size and necrosis rate of preoperative chemotherapy (P〉0.05). Conclusions XIAP plays an important role in the pathogenesis and development of osteosarcomas with the effect of inhibiting apoptosis. XIAP expression can be regarded as an independent risk factor in the evaluation of osteosarcoma's biological behavior.
