摘要
目的研究文冠木正丁醇提取物对大鼠佐剂性关节炎的影响并初步探讨其作用机理。方法用氟氏完全佐剂诱导大鼠产生佐剂性关节炎,以关节肿胀度为指标来观察文冠木正丁醇提取物对大鼠佐剂性关节炎的影响;研究文冠木正丁醇提取物对小鼠单核-巨噬细胞的吞噬功能、抗体生成水平及迟发型超敏反应的影响,初步考察其作用机理。结果文冠木正丁醇提取物对佐剂性关节炎大鼠的原发性和继发性关节肿胀均有抑制作用(P<0.05),且能改善大鼠的全身症状;文冠木正丁醇提取物2.0、1.0g/kg灌胃能显著抑制小鼠单核-巨噬细胞的吞噬功能、α,κ值均明显降低(P<0.05)、抑制绵羊红细胞诱导的小鼠抗体生成、抑制DNCB诱导的小鼠迟发型超敏反应(P<0.001)。结论文冠木正丁醇提取物对大鼠佐剂性关节炎有拮抗作用,其机理可能与抑制免疫功能有关。
Objective:To study the effects of n-Butanol Extract (BEX)of Xanthoceras Sorbifolia Bunge on rat model with adjuvant arthritis(AA) in rats and its mechanism .Methods: Rat model of adjuvant arthritis(AA) was induced by FCA. Hind paw swelling was observed to evaluate the effect of BEX and phagocytic function of monocyte-macrophage system, antibody level and delayed hypersensitivity were examined to explore its mechanism. Results: BEX can inhibit the primary and secondary hind paw edema and improve the general symptoms (P < 0.05). BEX (2.0,1.0 g/kg, ig) could markedly inhibit the phagocytic function of monocyte-macrophage system ,the delayed hypersensitivity induced by DNCB and the formation of antibody induced by sheep erythrocyte in mice(P < 0.05or P < 0.001).Conclusion: BEX of Xanthoceras Sorbifolia Bunge exerts a certain effect on adjuvant arthritis and its mechanism was probably related to the inhibition of immune function.
