反映成年大鼠脑组织神经发生水平显示方法的灵敏性和可靠性评价 被引量:12

摘要 目的评价反映成年大鼠脑组织神经发生水平的显示方法。方法采用免疫细胞化学法观察溴化脱氧尿核苷(BrdU)显示细胞增殖的可靠性、灵敏性,以及寻找BrdU给药的合适剂量。结果BrdU25mg/kg剂量组与50mg/kg、100mg/kg组相比,齿状回BrdU阳性细胞数差异明显(P<0.05),50mg/kg和100mg/kg两组无显著性差异(P>0.05)。BrdU25mg/kg剂量组阳性细胞不易与周围细胞区分;50mg/kg剂量组和100mg/kg剂量组阳性细胞较易识别。且BrdU阳性细胞大多数分化为神经元。结论BrdU标记法可以准确反映成年大鼠脑组织神经发生水平。 Objective To evaluate the experimental method of display the levels of neurogenesis in adult rats. Methods Immunohistochemistry were used to validate the reliability of BrdU labeling neurogenesis in DG and selected an appropriate dose of BrdU.Results The number of BrdU labeled cells in 25 mg/kg group were significantly lower than the other two groups(P< 0.05),and the labeled cells very lightly stained.The number of BrdU labeled cells in 50 mg/kg and 100 mg/kg weren't significantly difference (P >0.05),and the labeled cells were clearly labeled because darkly stained.The majority of BrdU labeled cells could expressed neuronal phenotype.Conclusion BrdU labeling method can displaying the levels of neurogenesis in adult brain.
