成年大鼠海马神经前体细胞的体外培养及诱导分化

摘要 目的 建立成年大鼠海马神经前体细胞的体外培养方法并探讨其生物学特性。方法 使用加入bFGF ( 2 0ng/ml)的无血清培养基培养成年大鼠海马神经前体细胞 ,MTT法检测细胞生长曲线 ,BrdU标记和免疫荧光化学方法观察培养细胞生物学特性。结果 加入bFGF培养的成年大鼠海马组织细胞可表达Nestin ,具有稳定连续的克隆增殖能力。经诱导分化后的细胞可表达神经元和胶质细胞的特异性标志物。结论 本实验分离的培养细胞具有可在体外稳定自我复制、大量增殖和多向分化潜能的特点 ,是理想的神经前体细胞。 Objective To establish an experimental method for in vitro culture of hippocampus neural precursor cells of adult rats,and to study adult neural precursor cells phenotypical.Methods The neural precursor cells from hippocampus of adult rats were in vitro cultured by using serum free medium added with bFGF(20 ng/ml).MTT,BrdU labelling method and immunofluorescence were respectivly utilized to detecte growth curves and phenotypical of adult neural precursor cells.Results The neural precursor cells from hippocampus of adult rats had the ability to contimuous proliferate and express Nestin.Furthermore, these neural precursor cells also expressed the marker of neuron and glial cell after induced differentiation.Conclusion In this experiment,our cultured cells which have ability to self replicate,proliferate and multi potent differentiate are adult neural precursor cells that we expect.
