摘要
目的 探讨补肾、扶正、益气补肾中药 ,对醋酸可的松所致肾虚老龄小鼠免疫功能的影响。方法 用醋酸可的松致老龄小鼠肾虚 ,经分别灌喂补肾、扶正和益气补肾方药后 ,用MTT比色法测定小鼠脾淋巴细胞的增殖 ;用胸腺细胞增殖法和ELISA法 ,观察脾淋巴细胞分泌IL 2和IL 12的水平 ;乳酸脱氢酶法检测脾脏NK细胞的活性 ;RT PCR法观察IL 2和IL 12mRNA的表达。结果 肾虚小鼠脾淋巴细胞的增殖活性 ,以及IL 2和IL 12的水平 ,均较正常小鼠明显下降(P <0 .0 5 ) ,两种细胞因子mRNA的表达也受到抑制。经补肾、扶正和益气补肾方药治疗后 ,以上检测指标均有不同程度的提高 ,其中益气补肾方药组提高最明显。
Aim To explore the effects of Chinese medicines of strengthening vital energy(SVE), tonifying kidney(TK) and tonifying kidney plus strengthening vital energy(TK+SVE) on immune function of aged mice with nephrasthenia caused by corticosterone. Methods Corticosterone was used to render aged mice nephrasthenia and the therapy was carried out respectively with Chinese medicines mentioned above via stomach perfusion. Lymphocytic proliferation, the levels of IL 2 and IL 12 production, and expressions of IL 2 and IL 12 mRNAs in spleen from the nephrasthenia mice were evaluated by MTT colorimetry, thymus cell proliferation assay, ELISA and RT PCR, respectively. Results The lymphocytic proliferation, the levels of IL 2 and IL 12 and their mRNA expressions in corticosterone group were significantly lower than those in normal mice. After treatment with three kinds of Chinese medicines above indexes were raised to a certain extent, of them, curative effect of TK plus SVE was optimal.Conclusion TK plus SVE probably is an ideal prescription of treating nephrasthenia.
