摘要
目的 探讨柯萨奇病毒 RNA阳性信号在克山病心肌细胞中的形态特征及病毒感染与发病的关系。方法 以 6 5例不同型别克山病尸检心肌组织蜡块为研究对象 ,用寡核苷酸探针与组织切片进行原位核酸杂交 ,以检测柯萨奇 B组和 A组病毒 RNA。结果 急型、亚急型及慢型克山病的阳性杂交信号检出率分别为 83.0 %、87.9%及 80 .0 %。阳性信号在急型、亚急型克山病心肌中颗粒粗大 ,境界较清晰 ,但分布比较局限 ;在慢型克山病中阳性信号多散在分布于各部位心肌细胞内 ,但颗粒较细小 ,境界不清。结论 心肌组织蜡块长期保存不影响杂交结果 ;吉林省各型克山病均与柯萨奇病毒感染密切相关 。
Objective To investigate the morphologic features of positive signal of Coxsackie virus RNA in the myocardium specimens of Keshan Disease(KD) and relationship between the virus infection and the occurrence of this disease.Methods With in situ hybridization, Coxsackie virus and its distribution were detected in 65 cases of KD (12 cases of acute, 33 subacute, and 20 chronic).Results The positive hybrid signal rates in acute, subacute and chronic KD were 83.0%?87.9% and 80.0%,respectively. Granules of positive hybrid signal were thick, and has clear border in the specimens of acute and subacute KD, but relatively localizedly distributed. In chronic cases, most granules of the positive hybrid signals within all ports of the myocardial cells which were small and without clear border.Conclusions It is feasible to perform the retrieval study on paraffin embedded myocardial tissues preserved for long term by using in situ hybridization method. There were close correlation between all kinds specimens of KD in Jilin province and coxsackie virus, and virus infection was one of the important components of multiple causality of KD.
