摘要 目的 研究几种常用促渗剂对来氟米特体外经皮渗透的影响。方法 采用水平扩散池 ,离体SD大鼠腹部皮肤用促渗剂预处理 12h ,以渗透速率为指标 ,用HPLC测定来氟米特的浓度。结果 月桂氮酮 (Azone)单独使用不能明显提高来氟米特的渗透速率 (P >0 .0 5 ) ,而丙二醇、油酸、Azone和丙二醇合用都能显著提高来氟米特的渗透速率 (P <0 .0 1)。结论 丙二醇、油酸、Azone和丙二醇的混合液 (1∶5 ) OBJECTIVE: To study the effects of several penetration enhancers on the permeation of leflunomide through skin in vitro. METHODS: Horizontal diffusion cell was adopted as the apparatus for in vitro skin permeation. Excised SD-rat abdomen skin was pretreated with penetration enhancers for 12 h. Using permeability as index, the concentrations of leflunomide in the samples were measured by HPLC. RESULTS: Compared with blank skin, exclusive use of Azone had little enhancement effect on the permeability of leflunomide (P > 0.05). Propylene glycol, oleic acid, Azone/propylene glycol (1:5) showed marked enhancement effect on the permeability of leflunomide (P < 0.01). CONCLUSIONS: Propylene glycol, oleic acid and Azone/ propylene glycol (1:5) were found to be the proper penetration enhancers in the transdermal preparations of leflunomide.
