端粒及端粒酶与细胞老化及年龄相关疾病 被引量:1

摘要 端粒及端粒酶参与细胞老化 ,调节细胞增殖与凋亡的动态平衡 ,端粒长度反映细胞的复制史 ,代表细胞的全部复制潜能 ,是细胞的“分裂时钟” (mitonicclock)。端粒随细胞的更新 (turnover) ,年龄性磨损 ,使端粒进行性缩短 ,当达到临界点 (hayflicklimitpoint)时细胞复制能力丧失 ,细胞老化。端粒酶与端粒长度的维持有关 ,防止端粒进行性缩短 ,阻止细胞衰老 ,抑制凋亡 ,为细胞增殖的分子标志 ,端粒、端粒酶与年龄相关疾病 (agedependentdiseases)如高血压、动脉粥样硬化、肿瘤、非胰岛素依赖性糖尿病等的发生发展有关。端粒起内到在定时机制 ,端粒的磨损不仅与老化有关 ,同时逐渐缩短的端粒使亚端粒区基因不稳 ,为复杂的多基因特征疾病的年龄依赖性提供满意的解释。
