对原发性高血压患者外周血和动脉组织中端粒酶活性的研究 被引量:5

摘要 目的 :探讨原发性高血压 (EH)患者外周血单个核细胞和胆囊动脉组织中端粒酶的活性。方法 :用端粒酶 PCR- ELISA方法测定 90例 EH患者和 56例血压正常者外周血端粒酶的活性 ,同时测定 2 4例 (EH、血压正常者各 1 2例 )胆囊动脉组织中端粒酶活性的变化 ,并与 2 1例结肠癌患者进行对比。结果 :EH组外周血单个核细胞的端粒酶活性比对照组明显增高 , 级、低危 EH患者中已有外周血单个核细胞端粒酶活性的增加 ,这种酶的变化并不随血压的水平和预后危险因素的增加而增加 ,但规则的降压治疗可使外周血单个核细胞端粒酶活性降低。EH患者动脉组织中端粒酶活性也比血压正常者和癌周组织高 ,但较癌组织中的端粒酶活性低。结论 :端粒酶是细胞增殖活性的分子水平标志物 ,外周血单个核细胞端粒酶的激活可能是 EH的危险因素之一 ,动脉组织中端粒酶活性的增加可能促进 Objective:To investigate the telomerase activities of peripheral blood mononuclear cells and biliary artery tissues of patients with essential hypertension. Method:The telomerase activity of peripheral blood mononuclear cells in 90 hypertensives and 56 normotensives was measured with telomerase PCR ELISA assay. Meanwhile the telomerase activities in biliary artery tissues of 12 hypertensives, 12 normotensives and 21 cases with colon carcinoma were also measured and compared. Result:The telomerase activity of peripheral blood mononuclear cells in hypertensive group was significantly greater than that in control group. Logistic regression study showed that the main influenced factors of hypertension were age, body mass index, blood insulin and telomerase activity, while the main influenced factors of the telomerase activity were hypertension and the number of lymphocyte. Further study showed that there had been increased telomerase activity in class one and low risk hypertensives, and it had not been increased with raised blood pressure and prognostic risk factors. But the telomerase activity was decreased with regular antihypertensive treatment. The telomerase activity in biliary artery tissues of hypertensive group was significantly higher than that in normotensive group and pericarcinoma tissues,but being much lower than that in carcinoma tissue.Conclusion:Telomerase was a mark of cellular proliferation activity in molecular level. The up regulation of telomerase activity in peripheral blood mononuclear cells might be one of risk factors in essential hypertension. The increased telomerase activities in artery tissures may play a role in hypertensive vascular remodeling.
