摘要
用反相液相色谱法测定了啤酒花浸膏中α -酸和β -酸含量 ,以NovapakC18(15 0mm× 3.9mmID ,4 μm)为分析柱 ,甲醇∶水 (85∶15 ,v/v ,磷酸调 pH =2 .5 )为流动相 ,紫外 313nm检测。样品用甲醇溶解过滤后直接进样 ,外标法定量。α -酸和β -酸的线性范围分别为 0 .0 4 2 5 - 0 .6 90mg/mL、0 .0 2 0 - 0 .32mg/mL ,平均加样回收率分别为 98.2 % ,97.3% ,RSD %分别为 1.3%、1.2 % (n =5 ) ,最低检测浓度均为 0 .0 0 1mg/mL。本法 7min完成一次分析 ,具有简单、快速。
A simple,rapid HPLC method has been developed for separation and determination of α-acid and β-acid in hop extract.The α-acid and β-acid were separated on a Novapak C 18 column (3.9 mm×150 mm,4 μm) with methanol-water (85∶15,v/v) orhophosphoric acid pH=2.5 as eluent.The flow rate was 1.5 mL/min.The detection was effected with UV detector at 313 nm.The separation of α-acid and β-acid was satisfied.A good linear relationship of α-acid and β-acid was found in the range 0.042 5-0.680 mg/mL and 0.020-0.320 mg/mL.The conefficients of varition of α-acid and β-acid standard were 1.3%,1.2%.The recoveries were 98.2% and 97.3%,respectively.The method is simple,rapid and accurate.
