摘要
目的 研究HLA DQAI基因与消化性溃疡的遗传关联。方法 用HLA基因的聚合酶链反应 限制性片段长度多态性核苷酸分型技术 ,以等位基因特异性的限制性内切酶 (ApalI、BsajI、HphI、FokI、MboⅡ、MnlI)消化HLA DQA1座位特异的PCR扩增产物 ,对来自武汉地区汉族人群中的 1 2 0例消化性溃疡 (50例胃溃疡、70例十二指肠溃疡 )和 50例正常人的HLA DQA1等位基因进行分析。结果 胃溃疡和十二指肠溃疡患者HLA DQA1 0 30 1基因频率较对照组显著性增高 (P <0 .0 5 ,且HLA DQA1 0 1 0 2基因频率较对照组显著性降低 (P <0 .0 5)。结论 HLA DQA1 0 30 1和HLA DQA1 0 1 0 2基因与消化性溃疡有一定的关联 。
Objective To study the genetic susceptibility of HLA DQA1 alleles to peptic ulcer.Methods We examined 120 patients with peptic ulcer (50 with gastric ulcer,70 with duodenal ulcer)and 50 healthy in controls were examined for HLA DQA1 genotypes from Wahan Hans. HLA DQA1 typing was carried out by digesting the locus specific polymerase chain reaction amplified products with alleles specific restriction endonuclease(PCR RFLP):Apa1 I,Bsaj I,Hph I,Fok I,MboⅡ,Mnl I.Results The allele frequcncy of DQA1 0301 was significantly higher in patients with gastric ulcer (64%)or duldenal ulcer (64.3%)than that in healthy controls (36%),In contrast,the allele frequency of DQA1 0102 was significantly lower in patients with gastric ulcer (8%) or duodenal ulcer (8.6%) than that in healthy controls (26%).Conclusions These findings showed that DQA1 0301 and DQA1 0102 were associated with peptic ulcer and that there were immunogenetic differences in the HLA DQA1 locus between peptic ulcer patients and healthy controls.
