摘要
设计桑树TrnL trnF基因间隔区序列的特异引物 ,扩增并分析了桑属育 71 1和桑莲 2个品种的TrnL trnF基因间隔区序列 ,其长度分别为 4 14bp和 4 17bp ,且富含A/T。该序列有 4 2 1个分析性状 ,具有 17个变异位点。在这些变异中主要以碱基的插入和缺失 (主要是插入 /缺失dA)为进化形式 ,其余的发生了少数置换。除这些变异位点以外的核苷酸序列完全相同 ,说明两者有高度同源性 ,与桑科、菊科和蔷薇科的TrnL trnF基因间隔区序列有一定同源性。用Clustalx软件对 2 1份材料的TrnL trnF基因间隔区序列进行聚类分析 。
The cpDNA TrnL trnF intergenic spacer sequence in the genus mulberry,Yu71 1 and Sanglian,were amplified and sequenced successfully,the size of which are about 414 and 417 base pairs,respectively.The spacer region are characterized by a AT rich fragment and the mean nucleotide compositions are 0.316 8(A) 0.334 5(T) 0.167 3(G) 0.191 4 (C).There were 17 mutations because of mainly indels between the TrnL trnF intergenic spacer of Yu71 1 and Sanglian and the sequence identity was 95.9%.Evolutionary relationships between these 2 and other 19 materials based on the TrnL trnF intergenic spacer in GenBank by cluster method,we found that genus mulberry shared high homology with family Moraceae,followed by family Rosebush and family Composite.These relationships are consistent with previous morphological groupings.
