摘要
用RAPD技术对桑属 1 2个种 3个变种的 4 4份材料和 1份构属材料的基因组DNA进行了多态性分析。在事先优化的反应条件下 ,用筛选的 2 4个随机引物扩增 ,共得到 1 1 3条清晰稳定的多态性片段 ,多态性达 72 0 % ,表明材料间有较丰富的遗传多样性。统计这些片段 ,根据扩增计算出遗传相似系数和遗传距离 ,然后用UPGMA法进行分析 ,构建了 4 5份材料间的系统发生树 ,并进一步探讨了材料间的亲缘关系。
mulberry accessions, which belong to 12 species and 3 varieties,and 1 cudrania were examined for RAPD analysis.Total 157 bands,which are approximately 100~2 500bp sizes,were produced from 44 mulberry accessions by using 24 10bp random primers selected from 182.Of them,113 bands showed polymorphism with the ratio of 72.0%.Based on RAPD data,Nei’s genetic similarity coefficients and genetic distance were calculated and analysied using the UPGMA dendogram.Additionally,phylogenetic relationship among 45 materials was determined.The results from cluster analysis indicated that they are basically consistent with the morphological classification.
