TGF-β受体及其信号转导分子Smad4在大鼠纤维化肝脏的表达及意义 被引量:9

摘要 目的 为了探讨TGF β受体及其信号转导分子Smad4在大鼠肝纤维化中的表达及作用。方法 采用原位杂交和免疫组化技术检测TGF β受体及其信号转导分子Smad4的表达。 结果 肝纤维化时TGF βⅡ和Ⅰ型受体及其信号转导分子Smad4的表达显著增加 ,Smad4的表达主要位于门脉系统纤维间隔和肝小叶之间的非实质细胞 ,而TGF βⅡ和Ⅰ型受体的表达主要位于汇管区、窦周和纤维间隔。另外 ,仅有少量肝细胞表达Ⅱ型受体。结论 随着肝纤维化的发展TGF β受体及其信号转导分子Smad4的表达明显增加 。 Objective To investigate the expression and the role of TGF β receptors and signal transduction molecule Smad4 in rat fibrotic liver.Methods In situ hybridization and immunohistochemistry was used.Results The expression Smad4 and the both TGF β type Ⅱand Ⅰreceptors were significantly increased in the fibrotic liver than in the control.The Smad4 mRNA and protein was mainly in nonparenchymal cells that distributed in the portal tract,fibrous septa, and the border between liver nodule and fibrous tissue.Also the TGF β type Ⅱ and Ⅰ receptor was mostly localized in the protal tract, around sinusoid and fibrous septa.In addition, only few hepatocytes expressed the type Ⅱ receptor.Conclusion The expression of the TGF β type Ⅱ and Ⅰ receptor and signal molecule Smad4 increase with the progress of liver fibrosis.It might be an improtant factor of fibrogenesis in fibrotic liver.
