磁共振单克隆抗体免疫靶向对比剂McAb-DMP的制备

摘要 目的 探讨磁共振单克隆抗体靶向对比剂McAb DMP的制备方法。方法 以葡聚糖为载体 ,采用氧化法制备免疫靶向磁性微粒。结果 制备的磁性纳米微粒与单抗相比保持了较高的结合活性和免疫活性。结论 合成CEA单克隆抗体标记磁性微粒 ,保证了结合活性免疫活性 。 Objective To explore a preparing method for MRI target contrast agent, dextran magnetite particles coated with CL 3 monoclonal antibody (McAb DMP). Methods With dextran as carrier, McAb DMP particles was prepared by periodate procedure. The immune activities of the prepared particles were determined with ELISA. Results McAb DMP particles had a higher binding activity with periodate sodium at an amount of substance ratio of 1∶2, and kept better immune activity compared with unbound McAb. Conclusion McAb DMP particles guarantee the binding and immune activity in compared with McAb, providing a basis for in vivo study on animals
