摘要
目的:建立固相萃取-高效液相色谱法测定血清中多西环素的方法。方法:美国Spectra-Physics高效液相色谱系统,色谱柱为Inertsil ODS-3。流动相为乙腈-水(50:50,v/v,含0.15%三氟乙酸),流速为1.0mL·min-1,柱温为室温,紫外检测波长为350nm。血清中加入内标土霉素,经C18固相萃取小柱提取后进样测定。结果:多西环素的线性范围为0.125~5.0mg·L-1,最低检出浓度为10μg·L-1。样本回收率为90.6%~104.3%。日内精密度<4.51%,日间精密度<5.30%。结论:本方法较为简便,结果准确可靠,适用于多西环素的药代动力学研究。
OBJECTIVE: To develop a method of determination of doxycycline in serum using high-performance liquid chromatography (HPLC), and a commercially available C18 solid phase extraction cartridge. METHODS: A Spectra- Physics HPLC system was used. The mobile phase was 50% (v/v) of acetonitrile in water containing 0.15% trifluoroacetic acid. Doxycycline and Oxytetracycline (internal standard) are extracted in the mobile phase and injected directly into the HPLC system. Doxycycline was separated on an Inertsil ODS-3 column and detected by an UV detector at 350nm..RESULTS: A good linear relationship was obtained for doxycycline concentration from 0.125 to 5.0 mg·L-1. The detection limit was 10μg·L-1 with an injection volume of 100μl.The recovery was 90.6%-104.3%. The intra-day RSD was less than 4.51% and the inter-day RSD less than 5.30%. CONCLUSION: The proposed method is sensitive, reliable and can be used for pharmacokinetic studies of doxycycline.
