摘要
目的:筛选及鉴定出血性中风相关基因。方法:应用差别显示聚合酶链反应(differential display PCR,DD-PCR)技术比较出血性中风病人脑组织出血部位及正常边缘部位的基因表达,并用RT-PCR研究其出血部位及周边正常部位基因的mRNA水平表达情况。结果:经DD-PCR筛选到17个差异片段,其中1个可能为出血性中风相关基因,即人类E2A/HLF融合蛋白mRNA,此基因经RT-PCR证实在出血性中风病人脑组织中高于正常组织。结论:E2A/HLF可能为出血性中风相关基因,在后者发病机制中有一定的作用。
Objective: To screen and identify the gene related to hemorrhagic stroke. Methods: Differential display polymerase chain reaction (DD-PCR) was used to compare genes expression between the part of control and the part of stroke in brain tissues of two patients, and the mRNA levels of the related gene were analyzed between them by the method of RT-PCR. Results: A human fusion protein expressed by E2A/HLF mRNA was found by the method of DD-PCR. RT-PCR confirmed that the level of E2A/HLF mRNA was higher in the part of stroke than that of control in brain tissue of stroke patients. Conclusions: E2A/HLF might be a stroke-related gene.
