摘要
目的:探讨血液系统恶性肿瘤病人化疗后应用rhG-CSF对中性粒细胞表型影响。方法:应用酶联免疫吸附法（ELISA）测定血浆rhG-CSF水平;应用免疫荧光技术和流式细胞仪分析检测中性粒细胞表型。结果:①急性白血病组和淋巴瘤组在应用rhG-CSF后的血浆rhG-CSF水平明显高于用药前的水平;②急性白血病组中性粒细胞CD64的表达明显高于正常对照组和感染组,在该组和淋巴瘤组中应用rhG-CSF后CD64的表达均较用药前有所增高;应用rhG-CSF前急性白血病组和淋巴瘤组CD62L的表达较正常对照组明显增高,而用药后其表达显著下调。急性白血病组和恶性淋巴瘤组应用rhG-CSF前、后,CD16、CD32、CD14和CD11b均无明显改变。结论:接受化疗的血液系统恶性肿瘤病人仍保留了对rhG-CSF的部分正常反应。
Objective: To explore the effect of recombinant human granulocyte colony-stimulating factor (rhG-CSF) on neutrophil phenotype in patients with hematological malignancies having undergone chemotherapy. Methods: The level of plasma rhG-CSF was detected by the method of ELISA. Neutrophil phenotype was analysed by flow cytom-etry using immunofluorescent techniques. Results:①After rhG-CSF administration, the level of plasma rhG-CSF was significantly increased in patients with acute leukemia and lymphoma. ② In the group of patients with acute leukemia, the neutrophils had a higher expression of CD64 than that in normal controls and patients with infection, and a mild increase of expression of CD64 was found in patients with acute leukemia and lymphoma during the treatment of rhG-CSF. Moreover, before injection of rhG-CSF, the group of patients with acute leukemia and lymphoma had a higher expression of CD62L than normal controls, and it was down-regulated after injection of rhG-CSF. No modifications of CD16, CD32, CD14 and CD11b were detected in these patients before and after injection of rhG-CSF. Conclusions: The patients with hematological malignancies having undergone chemotherapy have a partially normal response to rhG-CSF.
