诊断超声对人早孕绒毛细胞DNA影响的定量研究

摘要 目的 探讨诊断超声经腹照射子宫内孕囊对人早孕绒毛细胞DNA的影响。方法  60名拟行人流的早孕妇女随机分为照射 1 0min组和未照射组 ,各 3 0例。取绒毛组织后 ,以紫外分光光度法和3 2 P标记Alu探针打点杂交法定量检测绒毛细胞DNA单、双链断裂。结果 两种方法检测结果显示 ,照射组与对照组绒毛细胞单、双链DNA的量均无显著性差异 (P >0 .0 5 )。结论 诊断超声宫内照射孕囊 1 0min ,未引起人绒毛细胞DNA单、双链断裂 。 Objective To investigate the DNA damage in human early chorionic villus cells in uterus induced by diagnostic ultrasonography. Methods Sixty normal women in early pregnancy, who wanted to undergo artificial abortion, were randomly divided into two groups: exposure group of 30 cases exposed to diagnostic ultrasound through abdomen each for 10 minutes, 24 hours later chorionic villi were extracted; and none exposure control group with their chorionic villi also collected by curettage. Single stranded and double stranded breaks in DNA of villus cells in all cases were quantitated by spectrophotometry and using 32 P labeled Alu probe for dot blotting hybridization.Results No significant differences were shown in quantity in two parameters mentioned above between two groups ( P> 0.05 ). Conclusion The results indicated that in between exposed intensity and duration of diagnostic ultrasound, no single stranded breaks and double stranded breaks in DNA of human chorionic villus cells were observed, and no DNA damage was induced.
