摘要
在已建立的秦美猕猴桃叶片最佳再生系统的基础上 ,利用叶盘法和基因枪相结合的方法进行了秦美猕猴桃双价耐盐基因 m tl D/gut D的转化研究 ,经诱导不定芽及诱导生根阶段卡那霉素 (选择性抗生素 )连续筛选 ,获得了 2 0株卡那霉素抗性转化植株。抗性植株经 PCR检测 ,有 6株呈阳性。耐盐试验表明 4株阳性植株抗 Na-Cl能力比对照均有不同程度提高。 PCR及耐盐试验初步证明耐盐基因转化获得成功。
On the basis of the established Leaf explant regeneration system of kiwifruit (Qin mei),transformation of mtlD/gutD gene to kiwifruit (Qin mei) by means of leaf disc and microprojectile bombardment was undertaken for the first time.Through successive selection in the period of shoot and root induction under high level kanamycin pressure,20 kanamycin resistant rooted plants were obtained,out of which,6 were positive through PCR analysis.The salt resistant test of these 6 positive plants showed that 4 of them were enhanced in salt tolerance ability to some extent.PCR analysis and salt resistance test preliminarily indicated that the successful transformation has been made.
